We previously reported that Hwang Chan Go (HCG), composed of the major bioactiveconstituents of garlic, onion, and wild rice (Zizania latifolia), exerts therapeutic effects inanimal models of inflammatory disorders. In the present study, we hypothesized thatthese plants may also confer beneficial effects on hyperlipidemia, with particular emphasison the role of wild rice. To test this hypothesis, we evaluated the effects of alliin (a garlicconstituent), quercetin (an onion constituent), and zizania (a water extract of wild riceused as its constituent) on oleic acid (OA)-induced steatosis in HepG2 cells. To specificallyexamine the contribution of wild rice, zizania was tested alone or in combination withalliin, quercetin, or both. Cellular steatosis was assessed by quantifying lipid accumulationand by measuring the expression of free fatty acid receptor 1 (FFAR1), periostin (POSTN),3-hydroxy-methylglutaryl coenzyme A reductase (HMGCR), or peroxisome proliferatoractivatedreceptor alpha (PPARα), key regulators of triglyceride and cholesterol metabolism. The results showed that these compounds reduced lipid accumulation, downregulatedFFAR1, HMGCR, and POSTN, and upregulated PPARα. in OA-treated HepG2 cells. Notably,zizania potentiated the actions of alliin, quercetin, or their combination. These findingssuggest that zizania may have potential as a dietary supplement ingredient for managinghyperlipidemia and related metabolic disorders.
