Breast cancer is the most common malignant tumor among women, and early diagnosis of breast cancer can greatly improve patient survival rates and life quality. Therefore, the sensitive and accurate detection of the carbohydrate antigen 15–3 (CA15–3) as a marker of breast cancer is of great significance. In this work, luminol-europium nanocomposites (Lu-NCs) coordinated by luminol were used as novel electrochemiluminescence (ECL) luminophores for the immunodetection of CA15–3. Luminol is an aggregation-induced emission (AIE) molecule that self-assembles with Eu3+ to generate flower-like Lu-NCs, which produce more stable, stronger ECL signals and higher ECL efficiency than pure luminol. Subsequently, platinum-copper nanocages (PtCu NCs) with superoxide dismutase (SOD) and catalase (CAT) activities were used as quenchers to convert superoxide radicals (O2•−) into H2O2, which was further decomposed, leading to a reduction in the ECL signal. This system enabled the ultrasensitive detection of CA15–3 across a linear range of 0.001–100 U/mL, with a detection limit of 0.0004 U/mL. The successful development of this sandwich-type immunosensor enriches the luminescent aggregation induction and expands the application of mimetic enzymes in detection, providing a versatile platform for luminescent aggregation induction as well as the application of mimetic enzymes in cancer detection. © 2024 Elsevier B.V.
