Acute myeloid leukemia modeling based on human pluripotent stem cell-derived myelopoiesis A systematic review

Acute myeloid leukemia (AML) is a hematological malignancy attributed to clonal expansion of myeloid progenitors and impaired hematopoietic differentiation. Although conventional animal and cell line models are considered robust systems for clarifying pathological mechanisms, they may not fully recapitulate the molecular complexity and disease-associated phenotypes observed in patients with AML. Recently, pluripotent stem cells (PSCs) have emerged as a promising source to genetically engineer the patient-specific models. These PSCs can further differentiate into hematopoietic progenitors and myeloid lineages, thereby modeling key features of myeloid leukemogenesis in vitro. In the current review, we highlight recent applications of PSC-based leukemia modeling compared with those of conventional approaches and introduce the generation and characterization of PSC-derived myeloid cells. PSC-derived leukemia closely mimics primary patient-matched AML, enabling the precise dissection of clonal dynamics and the contribution of specific mutations to the disease phenotype. Accordingly, human PSC-based myelopoiesis platforms could serve as powerful tools to unravel pathogenic mechanisms and develop personalized medical approaches for patients with AML.
