Plain Language Summary: What is this summary about? This is a summary of the updated results of a study called PHAROS. The PHAROS study looked at the effects of a treatment in people with metastatic non-small cell lung cancer (NSCLC). The treatment in this study was a combination of two medicines called encorafenib and binimetinib (called ’encorafenib plus binimetinib’ in this summary). People in this study had a specific type of NSCLC with a change in a gene called BRAF in their cancer cells. This type of NSCLC is called BRAF V600E-mutant NSCLC. Since the first results of the PHAROS study, people in the study continued to be monitored over a longer period of time. These updated results looked at how well encorafenib plus binimetinib worked, and whether there were any side effects. What were the main findings from this study update? In this study, 98 people with BRAF V600E-mutant metastatic NSCLC were treated with encorafenib plus binimetinib. Before starting encorafenib plus binimetinib, 59 people had not received any previous treatment for their BRAF V600E-mutant metastatic NSCLC, and 39 people had received one previous treatment for their BRAF V600E-mutant metastatic NSCLC. At the time of this analysis, 50% of the people who had not received any previous treatment for their BRAF V600E-mutant metastatic NSCLC before receiving encorafenib plus binimetinib were expected to be alive for at least 47.6 months. And 50% of the people who had received previous treatment for their BRAF V600E-mutant metastatic NSCLC before receiving encorafenib plus binimetinib were expected to be alive for at least 22.7 months. The most common side effects of encorafenib plus binimetinib were nausea, diarrhea, fatigue, and vomiting. What do the findings from this study mean? These results from the updated analysis of the PHAROS study showed that people enrolled in the study continued to benefit from treatment with encorafenib plus binimetinib. People had similar side effects over the longer period of time compared to the first results of the PHAROS study. Clinical trial number: NCT03915951.
